چکیده فارسیعنوان : ارزیابی زیرگروه سلول B محیطی و بیان گیرنده ی BAFF/APRIL در بیماران کووید 19زمینه و هدف : ظهور بیماری کووید-19 و تاثیر گسترده آن بر سلامت جوامع بشری، لزوم انجام مطالعات در زمینه تاثیر این بیماری بر سیستم ایمنی بدن انسان ها برای کشف راه حل های موثر جهت مقابله با این بیماری را می طلبد. با توجه به نقش پاسخ ایمنی همورال در روند بیماری، همچنین عملکرد سایتوکاینهای BAFF و APRIL و رسپتور این سایتوکاینها در سطح لنفوسیتهای B، بر بروز کووید 19 و شدت بیماری، هدف از انجام این پژوهش بررسی جمعیت لنفوسیتهای B و میزان عرضه رسپتورهای BAFFR، TACI و BCMA در سطح این سلولها در بیماران کوويد ۱۹ میباشد.مواد و روش ها : مطالعه حاضر برای بررسی پاسخ ایمنی همورال در سال 1401 بر روی بیماران مبتلا به COVID-19 انجام شده است. در این مطالعه اطلاعات مربوط به 120 شرکت کننده (60 فرد بیمار مبتلا به کووید-19 و 60 فرد سالم کنترل) در طول 14 روز (روز اول، روز هفتم و روز چهاردهم) جمع آوری و به بررسی سوالات و فرضیات تحقیق پرداخته شده است. جداسازی سلولهای تک هسته‌ای خون محیطی با استفاده از فایکول و گرادیان شیب غلظت انجام گرفته است. پس از جداسازی PBMCs از خون محیطي بیماران و شمارش تعداد سلولهای زنده، بررسی فلوسایتومتریک سلولهای B و رسپتور های سطح سلولی BAFFR، TACI و BCMA طبق پروتوکل شرکت Biolegend.Co انجام شده است. یافته ها : میانگین سلول های BCD19 و همچنین میانگین سلول های B دارای رسپتور های TACI و BAFFR ،در گروه آزمون ،در روز هفتم پس از تشخیص به بیماری کووید 19 بیشتر از روز اول و روز چهاردهم میباشد و هرچند که میان تغییرات این متغیرها با جنسیت افراد شرکت کننده در مطالعه، ارتباط معنی داری دیده نمیشود اما در عین حال، تفاوت میانگین درصد سلولهای BCD19 بر حسب شدت بیماری، در روز هفتم معنادار میباشد به طوریکه میانگین درصد این سلولها در گروه اول (شدت بیماری کم) نسبت به گروه سوم (شدت بیماری شدید) افزایش معنادار داشته است.نتیجه گیری : یافته های مطالعه حاضر نشان داد که سطح سلول های BCD19 و سلول های BCD19 دارای رسپتور های TACI یا BAFFR در روز هفتم پس از تشخیص بیماری کووید 19 نسبت به روز اول تشخیص و روز چهاردهم، بیشتر میباشد و همچنین با افزایش شدت بیماری در روز هفتم، مقدار سلول های BCD19 کاهش یافته اند که مجموع موارد فوق، نشان دهنده ی پاسخ ایمنی هومورال ضعیف تر در فاز شدید بیماری میباشد. شاید بتوان گفت کاهش عملکرد پاسخ ایمنی همورال دلیلی بر وخامت حال بیماران میباشد. واژه های کلیدی: کووید -19 ، BAFF Receptors ، Humoral Immunity English AbstractIntroduction & Objective : The emergence of the covid-19 disease and its widespread impact on the health of human societies requires the need to conduct studies on the impact of this disease on the human immune system to discover effective solutions to deal with this disease. Considering the role of humoral immune response in the course of the disease, as well as the lack of sufficient studies on the role of BAFF and APRIL cytokines and the receptor of these cytokines on the level of peripheral blood mononuclear cells, on the incidence of Covid-19 and the severity of the disease, and due to the lack of a similar study in Iran, therefore, the purpose of this research is to investigate the supply of BAFFR, TACI and BCMA receptors on the surface of peripheral blood mononuclear cells in patients with Covid-19.Materials and Methods : The current study is a cross-sectional descriptive-analytical research that was conducted in 1401 on patients with COVID-19. In this study, information on 120 participants (60 patients with covid-19 and 60 healthy controls) was collected during 14 days (the first day, the seventh day, and the fourteenth day) and research questions and hypotheses were investigated. Isolation of peripheral blood mononuclear cells has been done using Ficoll and concentration gradient. After separating PBMCs from peripheral blood of patients and counting the number of live cells, a portion of the cell suspension was removed and for flow cytometric analysis of B cells and cell surface receptors BAFFR, TACI and BCMA according to the protocol of Biolegend.Co company and finally for analysis It has been used with a flow cytometry device.Result: This study shows that the average of BCD19 cells, as well as the average of B cells with, TACI and BAFFR receptors, in the test group, on the seventh day after contracting the disease of Covid-19 is higher than the first day and the fourteenth day, and also higher than the average is the control group and although there is no significant correlation with the gender of the participants in the study, but at the same time, the difference in the average percentage of BCD19 cells is significant according to the severity of the disease on the seventh day, so that on the seventh day after contracting the disease Covid-19, in patients who are in the first phase of their disease, the average number of BCD19 cells is higher than the second phase, and the second phase is higher than the third phase of the disease.Conclusion : In total, the findings of the present study showed that the level of BCD19 cells and BCD19 cells with TACI or BAFFR receptors is higher on the seventh day after contracting the covid-19 disease compared to the first day of infection and the fourteenth day, and also with an increase The severity of the disease on the seventh day, the amount of BCD19 cells increased, which indicates a more severe humoral immune response in the inflammatory phase of the disease.Keywords: B cell subset - BAFF/APRIL receptor - Covid 19
